pH imaging

Apoplastic pH in plants can be imaged using the Apo-pHusion biosensor (among others) expressed throughout the whole seedling (Gjetting et al., 2012). This allows for the observation of distinct acidic and basic pH zones along the seedling and enables tracking of apoplastic pH dynamics during plant growth, both in wild-type plants and in mutants affected in cellulose biosynthesis.

Calcium imaging

Chemical imaging is non-destructive and enables real-time, quantitative, and reliable measurement of chemical compounds. In this case, calcium imaging was performed using the RGeco-1 biosensor (Zhao et al., 2011), allowing for the detection and analysis of Ca²⁺ spikes in the root tip.
